A new research article has been published by the Pliotas group in the journal STAR Protocols (Cell Press). In the study entitled “HDX-guided EPR spectroscopy to interrogate membrane protein dynamics” we describe detailed protocols for an integrated EPR-based approach to study membrane protein dynamics, provide instructions for the sample preparation of spin-labeled membrane proteins using the mechanosensitive ion channel MscL as a model system, and use HDX mass spectrometry as a precursor to guide spin labeling strategies for EPR methods, such PELDOR (or DEER), ESEEM and CW-EPR spectroscopies.
